abstract = {The ITS regions of 10 species of Steinernema were PCR amplified using primers from flanking 18S and 26S sequences and directly sequenced using an automated sequencer. The sequences were aligned and used for species characterization and differentiation. Sequence length varied from 916 to 1061 base pairs (bp) in length. Restriction mapping of these sequences showed diagnostic variation such that the number of cuts and the length of the resulting fragments can be used for species differentiation. The composition and ratio of sequence similarity allow for further characterization of the ten nematode species. Phylogenetic relationships among the sampled Steinernema taxa were estimated by maximum parsimony and maximum-likelihood. Our results support the utility of the ITS region for differentiation of the nematode species of the genus Steinernema. The ITS region appears to be phylogenetically informative for closely related sister species, but high variability among more distantly related taxa preclude its use for resolving relationships among all members of the genus.}
