We were able to detect HSVd, but we were not able to detect CEVd in all collected samples. Thus, primer pairs, CEVd F194/R3, for successful detection of CEVd for Taiwanese isolates were developed. Additionally, we also developed multiplex RT-PCR for simultaneously detecting CEVd and HSVd using one-step RT-PCR. Real-time RT-PCR was also developed for more sensitive detection of CEVd and HSVd. Our field detection data demonstrated that although HSVd was only recently reported in Taiwan, it is more prevalent (30%) than CEVd (27.8%) and it is common to detect both CEVd and HSVd simultaneously in a citrus plant (18%).}

Abstract
Viroids cause diseases in citrus and have significant impact on the citrus industry. Two citrus viroids, CEVd (Citrus exocortis viroid) and HSVd (Hop stunt viroid) had been reported in Taiwan; however, the detection of these viroids still largely depends on bioassay and their prevalence and measurement for disease management in citrus remains to be investigated in Taiwan. In this study, we first collected samples from the field and used bioassay for viroid characterization, and also used primers (CEVd F1/R1 and HSVd F1/R1) reported by Bernad and Duran-Vila (2006) for RT-PCR detection. We were able to detect HSVd, but we were not able to detect CEVd in all collected samples. Thus, primer pairs, CEVd F194/R3, for successful detection of CEVd for Taiwanese isolates were developed. Additionally, we also developed multiplex RT-PCR for simultaneously detecting CEVd and HSVd using one-step RT-PCR. Real-time RT-PCR was also developed for more sensitive detection of CEVd and HSVd. Our field detection data demonstrated that although HSVd was only recently reported in Taiwan, it is more prevalent (30%) than CEVd (27.8%) and it is common to detect both CEVd and HSVd simultaneously in a citrus plant (18%).
