Abstract

Fomitopsis pinicola, Polyporus varius and Hymenopellis radicata are lignicolous fungi present in the Serbia region as well as in majority of forest ecosystems of northern hemisphere. Geographic variations in ITS1, 5.8S, ITS2 regions of these frequently occurring species were analyzed in this study. Sequences obtained from basidioms originated from different forest habitats in two distinctive parts of central Serbia (Mts Tara and Stara planina) were compared with sequences downloaded from publicly available NCBI and UNITE databases. The highest intraspecific variation within ITS1, 5.8S, ITS2 region was observed in H. radicata (5,9%) while sequences of P. varius and F. pinicola showed lower variability (3,9 and 3,4% respectively). Our results indicate that variability of ITS sequences are primarily dependent of fungal species and is generally lower within populations when compared with metapopulation variability. It was also determined that fungal species from wide geographic localities are genetically differentiated, indicating that ITS is suitable for phylogeographic investigation of the species F. pinicola, P. varius and H. radicata.
